One Round Amplification 3' Expression Target Labeling Using Ambion's MessageAmp™ II-Biotin Enhanced Labeling Kit up to 12 samples can be processed at one time. The recommended total RNA in-put is 1000ng with 100ng being the minimum. GEC staff will qc all samples prior to start of labeling the assays using the Agilent 2100 BioAnalyzer to verify sample integrity and the NanoDrop Spectrophotometer to verify sample concentration and purity. Please refer to the Guidelines for Sample Submission sheet prior to bringing your samples to the GEC. When you bring over your samples please fill out a sample drop off form, and insure your samples are legibly labeled with a sample name and your P.I.'s name.

Whole Transcript Target Labeling Service Using Affymetrix's GeneChip® Whole Transcript (WT) Sense Target Labeling kit up to 12 samples can be processed at one time. This assay is optimized specifically for use with the GeneChip® ST Arrays where "ST" stands for "Sense Target," and where the probes on the arrays have been selected to be distributed throughout the entire length of each transcript. This assay has two configurations with the key differences indicated in the following table.
Two Configurations of the WT Sense Target Labeling Assay (Affymetrix WT Sense Target Manual)
* rRNA Reduction is done by user. Affymetrix recommends RiboMinus Human/Mouse Transcriptome Isolation kit from Invitrogen.
Prokaryotic Labeling Please contact the GEC for pricing.
Affymetrix GeneChip® Hybridization Service 3' Expression Hybridization Service Following Affymetrix recommended protocols and procedures, biotin-labeled cRNA is hybridized to the appropriate GeneChip® overnight in the AFX HybOven480, post processed on the automated Fluidics450 Station, and scanned on the GC 3000 G7 Scanner. Data is extracted from the scanned image using AFX Expression Console software. An Excel spreadsheet of the extracted data is provided along with all files generated on the Affymetrix GeneChip® System. User is notified by e-mail that their data and project summary is available on the UWBC file server for their downloading convenience. Please refer to the Guidelines for Sample Submission sheet prior to bringing your samples to the GEC. When you bring over your samples please fill out a sample drop off form, and insure your samples are legibly labeled with a sample name and your P.I.'s name.
Researchers are responsible for purchasing all GeneChips®. Please refer to the current UW-Madison Academic Pricing Sheet for all GeneChip® prices. To ensure academic pricing, when ordering from Affymetrix (1-888-362-2447), please refer to UW Contract #10-5540.
Whole Transcript Sense Target Hybridization Service Following Affymetrix recommended protocols and procedures, end-terminus biotin labeled DNA is hybridized to the appropriate GeneChip® overnight in the AFX HybOven480, post processed on the automated Fluidics450 Station, and scanned on the GC 3000 G7 Scanner.
